What is Semax?
Semax is a synthetic peptide of 7 amino acids, based on a fragment of ACTH (adrenocorticotropic hormone) — amino acids 4-10 with an additional Pro-Gly-Pro tripeptide tail for stability.
It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences in the 80s. In Russia and some CIS countries, Semax is a registered drug for cognitive disorders and as a neuroprotector.
What is unique about Semax is that it retains the neurotropic properties of ACTH, but without the hormonal effects on the adrenal glands (it does not increase cortisol).
How does it work?
Semax modulates levels of brain-derived neurotrophic factor (BDNF) — a protein critical for neuron growth, survival, and plasticity. Increased BDNF is associated with improved memory and learning ability.
The peptide affects dopaminergic and serotonergic neurotransmission without acting as a direct agonist or antagonist. Instead, it modulates the tone of these systems.

A publication in Doklady Biological Sciences (2006) demonstrates that Semax affects the gene expression of more than 20 genes related to neuroplasticity and immune response in brain tissue.
