What is Melanotan 2?
Melanotan 2 (MT-2) is a synthetic cyclopeptide, an analog of α-MSH (alpha-melanocyte-stimulating hormone) — the natural hormone that controls skin pigmentation. It was developed at the University of Arizona in the 90s.
Natural α-MSH has a very short half-life (minutes), whereas Melanotan 2, due to its cyclic structure, is significantly more stable and more potent.
MT-2 activates the melanocortin receptors (MC1R-MC5R), which explains its multiple effects — not only on pigmentation, but also on appetite and other physiological processes.
How does it work?
Melanotan 2 binds to the MC1R receptor on melanocytes (the cells that produce melanin). Activation of MC1R stimulates the production of eumelanin — the dark pigment responsible for tanning.
Unlike UVB irradiation, which triggers tanning through DNA damage (and subsequent protective melanin production), MT-2 stimulates melanin synthesis directly, without requiring UV exposure.
MT-2 is a non-selective agonist — it also activates the MC3R, MC4R, and MC5R receptors, which explains its effects beyond pigmentation, including a potential reduction in appetite.
What does the research say?
A publication in the Journal of the American Academy of Dermatology (1999) demonstrated dose-dependent skin darkening in healthy volunteers receiving Melanotan 2 subcutaneously.
A study in Pigment Cell Research found that tanning induced by MT-2 increases the UV-protective properties of the skin, increasing melanin content without significant UV exposure.
A systematic review from 2020 summarizes the safety profile: the most common side effects include nausea (especially at the start), flushing, and changes in appetite. Most are mild and transient.

1. Broad-spectrum receptor activity — MT-2 is non-selective, activating multiple MC receptors. This explains its effects beyond pigmentation.
2. Individual variability of response — The pigmentation response varies significantly depending on phototype (per Fitzpatrick). Lighter phototypes respond more slowly.
3. Need for research — MT-2 is not approved by the EMA or FDA for cosmetic use. All data come from clinical and preclinical studies.
4. Gradual accumulation — The pigmentation effect is cumulative — it builds up gradually with repeated administration.
5. Photoprotection — Studies indicate that the increased melanin from MT-2 provides real UV protection, though it does not replace sunscreen.
