What Is IGF-1 LR3?

IGF-1 LR3 (Insulin-like Growth Factor 1, Long R3) is a synthetic analog of IGF-1 — one of the most important growth factors in the human body. The LR3 version is modified to achieve an extended half-life and increased biological effect.

Natural IGF-1 is produced mainly by the liver under the influence of growth hormone (GH). It mediates most of the anabolic effects of GH — muscle growth, bone mineralization, cell proliferation.

The LR3 modification involves substitution of an amino acid at position 3 (Glu→Arg) and the addition of 13 amino acids at the N-terminus. This reduces binding to IGF binding proteins (IGFBP), increasing the free fraction and biological activity.

How Does It Work?

IGF-1 LR3 binds to the IGF-1 receptor (IGF-1R) on the cell surface, activating the PI3K/Akt and MAPK/ERK signaling cascades. These pathways control cell growth, proliferation, and survival.

Unlike natural IGF-1, which quickly binds to IGFBP and becomes inactivated, the LR3 version remains free in the bloodstream for significantly longer. This extends the time window of biological activity.

IGF-1 LR3 stimulates hyperplasia (formation of new cells), not only hypertrophy (enlargement of existing ones). This characteristic distinguishes it from most anabolic agents.

What Does the Research Say?

IGF-1 is the subject of over 30,000 scientific publications on PubMed. Its role in cell growth, metabolism, and aging is among the best documented in endocrinology.

A study in the Journal of Biological Chemistry shows that the LR3 modification increases the biological potency of IGF-1 by an order of magnitude, due to reduced binding to IGFBP.

Research in the field of sports physiology demonstrates that IGF-1 plays a central role in muscle regeneration after injury and adaptation to physical exertion.

5 Key Facts About IGF-1 LR3

1. Extended half-life — The LR3 modification increases the half-life from ~15 minutes (natural IGF-1) to ~20-30 hours. This means more stable levels and less frequent administration.

2. Increased bioavailability — Reduced binding to IGFBP means more free IGF-1 at the receptor level.

3. Hyperplasia, not only hypertrophy — IGF-1 LR3 stimulates the formation of new cells, which is a unique mechanism compared to most growth factors.

4. Central role in the GH cascade — IGF-1 is the 'executor' of most GH effects. Its direct administration bypasses the need for GH stimulation.

5. Extensive scientific base — With tens of thousands of publications, IGF-1 is one of the most thoroughly studied growth factors in the history of endocrinology.
