What is GHRP-6?
GHRP-6 (Growth Hormone Releasing Peptide-6) is a synthetic hexapeptide, one of the first GHRP developed. It is a ghrelin mimetic with a pronounced effect on GH secretion and appetite.
Developed in the late 80s, GHRP-6 is the 'pioneer' of the GH secretagogue class. It is widely used in research settings and serves as a benchmark for comparison in the development of newer GHRP (Hexarelin, GHRP-2, Ipamorelin).
A characteristic feature of GHRP-6 is its strong stimulation of appetite — significantly more pronounced than GHRP-2 and Ipamorelin. This makes it suitable for research protocols where increased caloric intake is desired.
Mechanism of Action
GHRP-6 activates the GHS-R1a receptor in the pituitary gland, triggering the release of GH. At the same time, it suppresses somatostatin, removing the natural 'brake' on GH secretion.
It strongly activates the ghrelin system, which explains its pronounced effect on appetite. Ghrelin is the 'hunger hormone,' and its increase signals the brain that food intake is needed.
GHRP-6 also stimulates the secretion of IGF-1 from the liver — a secondary effect of increased GH, which mediates many of the anabolic effects of growth hormone.
What Do the Studies Say?
A study in Neuroendocrinology (1995) is one of the first to demonstrate that GHRP-6 elicits a powerful GH response upon intravenous administration in healthy volunteers.
A publication in Life Sciences (2006) examined the gastroprotective properties of GHRP-6 and found that the peptide may support the integrity of the gastric mucosa in animal models.
A study in Growth Hormone & IGF Research (2009) confirmed that the combination of GHRP-6 + GHRH produces a synergistic GH response exceeding the effect of either peptide alone.
