What is GHRP-2?
GHRP-2 (Growth Hormone Releasing Peptide-2) is a synthetic hexapeptide developed as a potent growth hormone stimulator. It acts as a ghrelin mimetic, binding to the GHS-R1a receptor in the pituitary gland.
It was developed in the early 90s and has been the subject of dozens of clinical studies in humans. It is considered the 'gold standard' within the GHRP class for balance between potency and side effects.
Unlike Ipamorelin, GHRP-2 also increases appetite through its ghrelin-like effect — which may be an advantage for people with insufficient caloric intake.
Mechanism of Action
GHRP-2 binds to the GHS-R1a receptor in the pituitary gland, triggering a cascade of intracellular signals (the IP3/DAG pathway) that lead to a massive release of GH from somatotropic cells.
At the same time, GHRP-2 suppresses somatostatin — the hormone that inhibits GH secretion. This dual mechanism (stimulation + removal of inhibition) explains why GHRP is so effective.
GHRP-2 also increases levels of ghrelin, IGF-1, and to a lesser degree — cortisol and prolactin. At standard doses, these increases are usually clinically insignificant.
What Does the Research Say?
A study in the Journal of Clinical Endocrinology and Metabolism (1997) showed that GHRP-2 induces a dose-dependent increase in GH in healthy men and women, with a peak increase of 5-10 times above baseline levels.
A publication in Endocrinology (2003) found that the combination of GHRP-2 + GHRH (CJC-1295) produces a synergistic effect — the GH response is significantly higher than the sum of the individual stimulations.
A 2019 meta-analysis concluded that GHRP-2 is an effective and well-tolerated GH secretagogue with a predictable response profile across multiple populations.
