What is DSIP?
DSIP (Delta Sleep Inducing Peptide) is a synthetic nonapeptide (9 amino acids), discovered in 1977 by the Swiss neurologist Grafton Schoenenberger. It was isolated from the brain tissue of sleeping rabbits and named for its ability to induce delta-wave (slow-wave) sleep activity.
Delta sleep is the deepest phase of sleep (NREM stage 3), during which most of the processes of physical recovery, growth hormone synthesis, and memory consolidation take place.
DSIP is naturally present in the brain and blood plasma of mammals, but its exact physiological role has not yet been fully clarified.
How does it work?
DSIP modulates the neurotransmitter systems associated with the sleep-wake cycle. Research indicates an influence on GABA, serotonergic, and dopaminergic neurotransmission.
In preclinical models, DSIP increases the duration of slow-wave sleep and reduces sleep latency (the time from lying down to falling asleep).
Interestingly, DSIP also demonstrates stress-modulating properties — it reduces corticosterone levels (a stress hormone in animals) and may support adaptation to chronic stress.
What does the research say?
The original publication in Pflügers Archiv (1977) describes the discovery of DSIP and demonstrates induction of delta-wave activity in rabbits following intraventricular administration.
A study in the European Journal of Pharmacology (1989) shows that DSIP normalizes sleep disturbances in rats under stress conditions, without causing daytime sleepiness.
A clinical observational study from the 1990s notes an improvement in subjective sleep quality in patients with chronic insomnia receiving DSIP. However, larger controlled trials are needed.
