What is Retatrutide?

Retatrutide is an experimental next-generation triple agonist that simultaneously activates three receptors: GLP-1, GIP and the glucagon receptor. This makes it the first compound to target all three key pathways of metabolic regulation.

It is currently in phase 2–3 clinical trials. Early results identify it as potentially the most effective weight-loss agent developed to date.

How it works

GLP-1 activation: suppresses appetite, slows gastric emptying and improves insulin secretion.

GIP activation: improves fat metabolism and insulin sensitivity, with a synergistic effect alongside GLP-1.

Glucagon activation: the unique third mechanism that increases energy expenditure (thermogenesis), stimulates the breakdown of fat stores in the liver and improves the lipid profile.

The triple combination addresses obesity on three fronts: reduced intake, improved metabolism and increased expenditure.

Clinical results

In the phase 2 study (48 weeks, 338 participants), retatrutide 12 mg achieved an average loss of 24.2% of body weight, the highest recorded value for a pharmacological intervention.

100% of participants at the maximum dose lost at least 5% of their weight. More than 90% lost over 10%.

Significant improvement in hepatic steatosis (fatty liver), a unique advantage attributed to glucagon activation.

How it is administered

A subcutaneous injection once a week. Starting dose: 2.5 mg/week, with gradual titration to 6–12 mg according to tolerance.

5 mg pen: 17 µg per unit. 10 mg pen: 33.3 µg per unit. 15 mg pen: 50 µg per unit.
